چکیده

Abstract Background DHA‐H (2‐hydroxy‐docosahexaenoic acid) is a promising therapeutic approach for Alzheimer’s Disease (AD). gives rise to dose‐dependent increased levels of HPA (Heneicosapentaenoic in blood plasma and brain whereas remains virtually absent from the DHA‐H‐treated mice. Oral administration transgenic model AD (5xFAD mice) induces significative cognitive recovery. However, molecular mechanism underlying this neuroprotective effect largely unresolved. Method Screening potential receptors was performed using computational tools. PPARG (Peroxisome Proliferator‐Activated Receptor γ) activity determined by cell‐based assays. Levels proteins involved amyloid production as well synaptic markers were WB and/or ELISA. β‐ γ‐secretase activities measured cell‐free Neuronal density immunohistochemistry. Brain Electrospray Ionization – Mass Spectrometry (ESI‐MS). Result Both are activators PPARG. We have analyzed treatment on amyloidogenic route 5xFAD mice related with activity. observed modulation protein enzymatic β‐amyloid prevention neuron degeneration compared untreated controls. These changes improved scores these Conclusion All results together indicate that must prevent neuronal decline through its immediate metabolite without apparent involvement

Peroxisome proliferator-activated receptor-γ protects against vascular aging.

Vascular disease occurs commonly during aging. Carotid artery and cerebrovascular disease are major causes of stroke and contributors to dementia. Recent evidence suggests that peroxisome proliferator-activated receptor-γ (PPARγ) may play a protective role in the vasculature, but the potential importance of PPARγ in vascular aging is unknown. To examine the hypothesis that PPARγ normally protec...

Truncated peroxisome proliferator-activated receptor-γ coactivator 1α splice variant is severely altered in Huntington's disease.

BACKGROUND Reduced peroxisome proliferator-activated receptor-γ coactivator 1α (PGC1α) gene expression has been observed in striatal cell lines, transgenic mouse models of Huntington's disease (HD), and brain tissue from HD patients. As this protein is a key transcription regulator of the expression of many mitochondrial proteins, these observations strongly support the role of aberrant mitocho...

Peroxisome Proliferator-Activated Receptor-γ Is Critical to Cardiac Fibrosis

Peroxisome proliferator-activated receptor-γ (PPARγ) is a ligand-activated transcription factor belonging to the nuclear receptor superfamily, which plays a central role in regulating lipid and glucose metabolism. However, accumulating evidence demonstrates that PPARγ agonists have potential to reduce inflammation, influence the balance of immune cells, suppress oxidative stress, and improve en...

Intestinal antiinflammatory effect of 5-aminosalicylic acid is dependent on peroxisome proliferator–activated receptor-γ

5-aminosalicylic acid (5-ASA) is an antiinflammatory drug widely used in the treatment of inflammatory bowel diseases. It is known to inhibit the production of cytokines and inflammatory mediators, but the mechanism underlying the intestinal effects of 5-ASA remains unknown. Based on the common activities of peroxisome proliferator-activated receptor-gamma (PPAR-gamma) ligands and 5-ASA, we hyp...
